Are you referring to the command line C compiler, et. al.? For as long as I've been using Visual Studio on Windows (at the Command Prompt), I've had to run the VCVARS[32|64].BAT batch file to set up the "DOS environment". For me, they live here:
C:\Program Files (x86)\Microsoft Visual Studio 10.0\VC\bin\vcvars32.batC:\Program Files (x86)\Microsoft Visual Studio 10.0\VC\bin\amd64\vcvars64.bat
